In vitro micropropagation of grayish oak (Quercus pedunculiflora K. Koch.) in normaland experimental water stress conditions

Quercus pedunculiflorahas been propagated in vitrofrom nodal segments of seedling 4months of age, green and mature acorns. The nutrient media, Woody Plant Medium (WPM) and Gres-soff and Doy, suplimented with benzylaminopurine (BAP 0,8 mg/l or 1 mg/l) have stimulated well theinduction of auxillary and adventitious buds. The development of induced buds into shoots occurredon nutrient medium supplemented with a low concentration of BAP (0,8 mg/l). The optimum con-centrations of mannitol or polyetylenglycol that can be achieved in vitroselection of the grayish oakshoots have been situated between 12 - 15 g%.Roots were formed on shoots placed in half - reduced salts medium, containing lowconcentration of auxins (IBA 0,3 mg/l) and NAA 0,1 mg/l) in 4-5 weeks. Rooted plants weretransplanted into pots containing a mixture of soil and perlite (1o:1 v/v) for acclimation.References
